Description
  The SureSilencing™ shRNA Plasmids are designed to specifically knock down the expression of individual genes by RNA interference under either transient (with GFP) or stable transfection (for hygromycin, neomycin or puromycin-resistance) conditions after performance of the appropriate enrichment or selection procedures, respectively. Each vector contains the shRNA under control of the U1 promoter and either the GFP gene, for the enrichment of transiently transfected cells, or the neomycin or puromycin resistance genes, for the selection of stably transfected cells.

Protocol
  First time users, please refer to the complete protocol in the SureSilencing™ shRNA Plasmid User Manual.
  1. Transform 1 µl provided plasmids separately into 50 µl competent E. coli cells according to manufacturer's instructions.
  2. Pick colony for large-scale overnight bacterial culture.
  3. Prepare stock of high-quality transfection-grade plasmid using a purification kit according to manufacturer's instructions.
  4. Transfect plasmids separately into replicate wells of your cell line of interest using conditions previously optimized for the transfection of plasmids into your cell of interest.
  5. Enrich the transfected cell population by FACS for GFP or by selection for neomycin or puromycin resistance.
  6. Assay the extent of knock-down by real-time RT-PCR in triplicate for each transfection.
  7. Perform your biochemical, cell or molecular biological gene function assay.

Kit Contents:
Pre-designed shRNA Plasmids include four tubes each containing individual shRNA Plasmids specific for the same gene. A fifth tube contains a negative control shRNA Plasmid. All plasmids provided are at concentration of 30 ng/µl - 50 ng/µl
        Catalog Numbers ending in "G" contain plasmids with the GFP marker.
        Catalog Numbers ending in "H" contain plasmids with Hygromycin-resistance marker.
        Catalog Numbers ending in "N" contain plasmids with the Neomycin-resistance marker.
        Catalog Numbers ending in "P" contain plasmids with the Puromycin-resistance marker.

Storage Conditions: The SureSilencing™ shRNA Plasmids are shipped on dry ice or cold packs. Store all tubes at -20 ºC.
